Abstrakt:Complex gene clusters cover about 5% of human genome. They are characteristic for multiple repetitions and this matter of fact complicates their correct assembly. Main goal of this study is to introduce correct process for gene clusters assembly and methods for later observation of final assemblies. We approach to a problem of bacterial artificial chromosomes (BAC) sequencing and subsequent assembly without any reference genome and apply the knowledge to assembly of the input BACs. Focusing on dot–plot matrix as adequate method to visualise structure of gene clusters, we discussed their pattern and illustrate various events in the sequence of gene clusters. We compare our final assemblies of various primates gene clusters to available reference genomes and make an outline from structural and also evolution perspective. In this study, we work with UGT2 gene cluster of seven primate species: Colobus guereza, Gorilla gorilla gorilla, Chlorocebus aethiops, Eulemur macaco, Lemur catta, Plecturocebus moloch, Ateles geoffroyi. First we compare assembled gorilla UGT2 gene cluster against three available reference gorilla genomes. As second task we do a comparison of all done UGT2 gene cluster assemblies with human reference of the gene cluster and study evolution history between species and in the gene cluster itself.
